
Ceftazidime + Avibactam
ZAVIALFA is a fixed-dose combination of ceftazidime, a cephalosporin antibiotic, and avibactam, a β-lactamase inhibitor. Ceftazidime disrupts bacterial cell wall synthesis by binding to penicillin-binding proteins, causing cell lysis. Avibactam protects ceftazidime from degradation by β-lactamases, including those from ESBL- and carbapenem-resistant bacteria. Effective against 95% of Pseudomonas aeruginosa and over 99% of enterobacteria, ZAVIALFA is a first-choice treatment for resistant gram-negative infections. It has a longer half-life of 2.7 hours compared to other β-lactamase inhibitors.

MODE OF ACTION:
Zavialfa is a fixed-dose combination medication consisting of ceftazidime, a cephalosporin antibiotic, and avibactam, a β-lactamase inhibitor. Ceftazidime works by binding to penicillin-binding proteins (PBPs) on the bacterial cell wall, inhibiting PBPs involved in the assembly and remodeling of the cell wall. This disruption prevents the cross-linking of peptidoglycan chains, weakening the bacterial cell wall, leading to cell lysis and bacterial death. Avibactam, a non-β-lactam β-lactamase inhibitor, inactivates certain β-lactamases (including Ambler class A, C, and some class D β-lactamases) via a unique, reversible covalent binding mechanism. This protects ceftazidime from degradation by β-lactamases and restores its activity against ceftazidime-resistant, β-lactamase-producing bacteria, without affecting its activity against susceptible organisms. Together, this combination enhances the effectiveness of ceftazidime, enabling it to combat resistant bacteria more effectively.
INDICATIONS
- Complicated Urinary tract infections
- Complicated Intra-abdominal infections
- Hospital-acquired pneumonia (HAP)
- Ventilator-associated pneumonia (VAP)
Recommended Adult Dosage
Zavialfa 2.5g is administered every 8 hours for:
- 7 to 14 days for HAP/VAP and complicated UTIs
- 5 to 14 days for complicated IAI
