Faropenem

Faropenem is a novel oral carbapenem antibiotic belonging to the penem group. It is a broader-spectrum antibiotic that is 4-8 times more active against methicillin-susceptible Staphylococcus aureus (MSSA) compared to other commonly used antibiotics. Faropenem offers a significant advantage over other parenteral carbapenems, as it is available in an oral form, making it more convenient for patients. It demonstrates lower minimum inhibitory concentrations (MICs) than other antibiotics like penicillin, ampicillin, amoxicillin, and cefuroxime, making it effective at lower doses. Faropenem is used to treat a wide range of bacterial infections, including those of the urinary tract, respiratory tract, skin, ears, nose, and throat, as well as gynecological infections.

MODE OF ACTION:

Faropenem is a beta-lactam antibiotic that exerts its bactericidal effect by targeting the bacterial cell wall, which is essential for the structural integrity and survival of bacteria. The cell wall provides a protective barrier, preventing the cell from collapsing under osmotic pressure and maintaining its shape. Without a functional cell wall, bacteria would be unable to maintain their structural integrity, leading to cell death.

Faropenem specifically interferes with the process of cell wall biosynthesis by binding to penicillin-binding proteins (PBPs). PBPs are a group of enzymes found in bacterial cell membranes that play a crucial role in the final stages of the peptidoglycan layer synthesis—the main component of the bacterial cell wall. This process involves cross-linking the peptidoglycan chains to form a rigid and stable wall.

By binding to PBPs, Faropenem inhibits the cross-linking process, thus preventing the completion of the cell wall formation. This results in a weakened cell wall that is no longer able to provide the necessary protection to the bacterial cell. As a consequence, the bacterial cell becomes unstable and more vulnerable to osmotic pressure, which eventually causes the cell to burst (a process known as lysis). The disruption of the bacterial cell walls and the resulting cell lysis leads to the death of the bacteria. This bactericidal action eliminates the bacterial pathogen, effectively treating the infection and preventing further bacterial growth or reproduction.
